Abstract
The objective of this study is to determine the effects of different type of combined administration with vitamin K:> (MK<|) and bisphosphonate (risedronate) on the bone mineral density, trabecular bone structure, and bone strength. 37, nine week-old female ICR mice were used for this experiment. After they were kept with regular feed for 30 days, the test mice were divided into five groups, based on the administration methods of risedronate(R) and MK.)(K) as follows; 1) sham 2) OVX-control (control), 3) concomitant administration of R and K (2RK), 4) administration of K after R (R-+K), 5) administration of R after K (K->R). After 16-week observation period, the left femur of each mouse was removed and subjected to analysis of bone mineral density, bone structure by bone peripheral quantitative computed tomography (pQCT) and micro focus x-ray CT(,uCT).
In the pQCT examination, the total bone mineral density in K-"R group showed significantly higher values than the control group. The u CT analysis revealed that the trabecular bone structure in K->R group displayed a significant improvement, compared with the control group. In the mechanical test for bone strength, only K->R group showed a significantly higher improvement in comparison to the OVX-control group. It was suggested that sequential administration of vitamin KĄ and risedronate was superior to other types of combined administration for treating osteoporosis.
